Missions

The project aims at developing foldamer-based approaches for mimicking secondary structures such as α-helices and for inhibiting protein-protein interactions (PPIs) with a focus on therapeutically relevant targets. The ability to design and synthesize sequence-specific synthetic oligomers that fold into well-defined secondary structures (i.e. foldamers) akin to those found in proteins opens up enticing opportunities for mimicking peptides and addressing some of their limitations such as short in vivo half-lives and poor membrane permeability. In particular, many foldamer backbones are endowed with useful properties such as structure predictability and improved resistance to enzymatic degradation that make them well suited as new modalities for therapeutic applications.

Work Context

CBMN and IECB are dynamic interdisciplinary research centers with technology platforms bringing the state-of-the-art scientific instrumentation and expertise in biophysical techniques, structural biology as well as preparative and analytical techniques. The project is interdisciplinary and will involve collaboration with laboratories at other Universities with opportunities for secondments.

The initial contract is for 12 months and can prolonged for another 12 months.

Constraints and risks

Experiments related to chemical synthesis will be carried out in the laboratory space of the PI within the European Institute of chemistry and Biology (IECB) a properly equipped and maintained laboratoty.

Missions

A 12-month postdoctoral contract, renewable for 12 months, is available at the Institute of Chemistry and Biology of Membranes and Nanoobjects located in Pessac as of September 2019. The project is linked to a CNRS contract (CNRS Momentum) and involves the detailed characterization of protein/membrane systems by solid-state Nuclear Magnetic Resonance (NMR) spectroscopy. The host laboratory is located in the premises of the European Institute of Chemistry and Biology in Pessac. The project's research program focuses on the structural study of these protein/lipid assemblies using solid NMR methods and identifying protein/lipid membrane interactions.

It hosts 15 research teams and has a very large number of structural biophysics platforms (NMR, X-ray, MS, Vibration, AFM, etc.). The postdoctoral student's work will consist of producing the system and recording and analyzing the solid-state NMR experiments. This work will be performed in the team "NMR of Membranes and Protein Assemblies". The team has access to NMR spectrometers ranging from 300 to 800 MHz, equipped with solid-state NMR probes.

Description of the thesis topic

The formation of co-crystals (molecular assembly of two species) with compressed CO2 emerged only very recently. The objective of the thesis is to go above the proof of concept by focusing on time-related events that should control the nature of the produced crystals and the purity of the produced powders in cocrystals, two characteristics of primary importance for the targeted pharmaceutical applications. To address such kinetic aspects, the crystallization will be first studied in an innovative high-pressure CO2 microfluidic plateform where in-situ diagnostic will be implemented before scaling up to larger set-ups. The crystallization will be then applied to chiral resolution.

Work Context

The candidate will work in a dynamic environment thanks to the co-working between CBMN that has the background in CO2-cocrystals and ICMCB's team specialist of microfluidic, and thanks to the framework of a national project that gathers specialists of conventional crystallization including with chiral molecules.
